Semax vs Selank: Two Cousins, Two Different Targets

Any semax vs selank comparison starts from a shared origin: both come out of the same Russian institute and both use the same route of administration. They act on different systems, though. Semax works on BDNF and dopaminergic systems, oriented toward activation and attention; Selank works on GABAergic and serotonergic systems, oriented toward reducing anxiety.

The shared origin

Both come from the same line of Soviet and later Russian research on regulatory peptides: short molecules derived from larger proteins, modified to resist degradation and retain activity on the central nervous system.

They also share:

  • Origin as a fragment of a larger molecule: ACTH in the case of Semax, tuftsin in the case of Selank.
  • Modification with proline and glycine to increase stability.
  • The intranasal route as the usual form of administration.
  • Registration as a medicine in Russia, with no approval in other frameworks.

What they do not share is the mechanism, and therefore not the intended effect either.

When each one is studied

Within the Russian registration, their indications do not overlap:

Semax has been studied in settings where the goal is to sustain or recover cognitive and attentional function.

Selank has been studied in anxiety settings, where the goal is to reduce reactivity.

These are opposite directions along the same axis. They are not interchangeable, and reading semax vs selank as "two nootropics" loses exactly what separates them.

The combination: what supports the hypothesis, and what evidence does not

The combination of the two circulates, with an argument worth examining.

The hypothesis: they act on different systems and in complementary directions (cognitive activation without the anxious reactivity that sometimes comes with it). Since they do not compete for the same receptor, interference would be unlikely.

What is missing: no published study evaluates the combination. Not preclinical, not clinical. The same reasoning applies here as with the BPC-157 and TB-500 combination: a coherent mechanistic hypothesis is not a finding, and a combination can behave differently from the sum of its parts.

Without a three-arm design (A alone, B alone, A+B), there is no way to know whether the effect is additive, synergistic, null, or interfering.

Differences in tolerability profile

Both are described with favorable profiles in Russian clinical use, with infrequent adverse effects, largely related to local irritation from the intranasal route.

The differences described:

Semax, given its activating orientation, has been associated with disturbed sleep when it is administered in the second half of the day.

Selank, despite being described as minimally sedating, acts on the GABAergic system, which makes caution reasonable regarding interactions with other substances that act on that system.

Neither observation comes from tolerability studies accessible for outside evaluation.

What the evidence does not settle

For both, and for the combination:

  • Efficacy under Western standards: no trials that meet them.
  • Human pharmacokinetics by the intranasal route: not quantified in accessible literature.
  • Prolonged use and tolerance: no data.
  • The combination: no studies at all.

The frame is the same one that applies to this whole cluster: promising research, not proven under the standards treated here as the reference.

Which one is better?

The semax vs selank question has no answer as posed, because the two are not after the same thing. Semax points toward activation and attention; Selank, toward reduced anxiety. Those are different directions.

Can they be used together?

That practice circulates, and its rationale is mechanistic: they act on different systems and do not compete for the same receptor. There is no study of the combination.

Is either one approved outside Russia?

Neither. Both are registered in Russia for certain indications and lack approval from the FDA, EMA, and INVIMA.

Why are they given through the nose?
